Tomorrow, November 20, we are booked to take the overnight Willer bus from Tokyo to Hiroshima. My question is, do we have to board at Shinjuku Station and definitely get off at Hiroshima?? Ideally, we'd like to board closer to Mishima and get off closer to Mihara, since those are the places we are and want to go...

I've taken these buses. usually you can hop on or drop off in any of its stops, they won't stop you, just let them know.
But its better to buy the correct ticket for wht you need because it may be cheaper
